After oral administration, the highest levels of antidepressant in the bloodstream occur between about 2-6 hours. Most tricyclics have a half-life of about 24 hours, which makes daily dosing possible. Levels decrease as the drug is metabolized by the liver into compounds with similar pharmacological properties. By the time the drug leaves the body, less than 5% remains unchanged.
